Observation of structural dynamics of meta-aggregates

The process of meta-aggregate formation of amyloid proteins, which is associated with neurodegenerative diseases, remains veiled due to the coexistence of diverse aggregate species. On the other hand, the advent of high-speed atomic force microscopy (high-speed AFM) has made it possible to image the structure and temporal changes of all biological macromolecules within the observation field of view at the single molecule level in liquid with nanometer spatial resolution and subsecond temporal resolution. This feature of high-speed AFM allows us to reveal the structural dynamics of each and every aggregate during the meta-aggregate formation process. In this study, we will clarify the meta-aggregate formation process of various amyloid proteins and the effect of drugs that interfere with structural dynamics of meta-aggregate formation.
